How does Fermachanneling work?


Fermachanneling works through manipulation of mutB to improve erythromycin production for strains and processes that are limited by ketide precursors. This includes the wild type strains of these organisms and could include commercial strains whose limiting factors are unknown or known to be ketide limited.


In carbohydrate-based medium mutB knockouts are used to boost production (Reeves et al., 2006; PDF).  In an oil-based medium, mutB operon duplications are used to accomplish the same goal (Reeves et al., 2007; PDF).


What are the Fermachanneling related products?


For Saccharopolyspora erythraea the channel-blocker for carbohydrate-based fermentations is the gene replacement plasmid pFL2132.  The channel-booster for  oil-based fermentations is the single crossover insertion plasmid pFL2212.


For Aeromicrobium erythreum the channel blocker for carbohydrate-based fermentations is the single crossover knockout plasmid pFL2106. There is no channel-booster for A. erythreum since this strain does not grow in oil-based fermentations.


What is the Dmr-phenotype technology?


“Dmr-phenotype” is strain improvement technology for mutB strains of S. erythraea (Weber et al., 2011; PDF). When the mutB strain is grown in the presence of 15 mM diethyl methylmalonate, it has been shown to produce 30% more erythromycin than the parent strain with, or without supplementation. The implications from Dmr phenotype research are that other mutations, besides mutB, can be added to commercial strains to boost their deoxysugar production. Once found, these mutants can be supplemented with diethyl methylmalonate to improve production of erythromycin A. A simplified method for finding Dmr-phenotype related mutations is described in the paper by Weber et al., 2011 (PDF).
